Stressed-out tadpoles grow larger tails to escape predators

Mar. 5, 2013 ? When people or animals are thrust into threatening situations such as combat or attack by a predator, stress hormones are released to help prepare the organism to defend itself or to rapidly escape from danger -- the so-called fight-or-flight response.

Now University of Michigan researchers have demonstrated for the first time that stress hormones are also responsible for altering the body shape of developing animals, in this case the humble tadpole, so they are better equipped to survive predator attacks.

Through a series of experiments conducted at field sites and in the laboratory, U-M researchers demonstrated that prolonged exposure to a stress hormone enabled tadpoles to increase the size of their tails, which improved their ability to avoid lethal predator attacks.

"This is the first clear demonstration that a stress hormone produced by the animal can actually cause a morphological change, a change in body shape, that improves their survival in the presence of lethal predators. It's a survival response," said Robert Denver, a professor of molecular, cellular and developmental biology and of ecology and evolutionary biology.

The team's surprising findings are detailed in a paper to be published online March 5 in the journal Proceedings of the Royal Society B. First author of the paper is Jessica Middlemis Maher, a former U-M doctoral student, now at Michigan State University, who conducted the work for her dissertation.

Scientists have long known that environmental changes can prompt animals and plants to alter their morphology and physiology, as well as the timing of developmental events. For example, tadpoles can accelerate metamorphosis into frogs in response to a drying pond, a high density of predators or a lack of food.

The term "phenotypic plasticity" is used to describe modifications by animals and plants in response to a changing environment.

"There's been a lot of interest in phenotypic plasticity among developmental biologists and evolutionary ecologists for more than 70 years, but there's been relatively little focus on the mechanisms by which the environmental signal is translated into a functional response," Denver said.

"We've known, for example, that tadpoles can change their body shape in response to predation risk. But until now, nobody knew the basic physiological mechanisms mediating that response. That's what's novel about this study."

The study involved wood frog tadpoles and the stress hormone corticosterone, which is similar to the human stress hormone cortisol. Tadpoles were collected from ponds at U-M's E.S. George Reserve in Pinckney, Mich., northwest of Ann Arbor.

Some of the tadpoles were reared in tanks at the reserve. Dragonfly larvae, which are known predators of tadpoles, were placed in small cages inside the tanks and were fed live tadpoles. When under attack, tadpoles release chemical signals called pheromones that travel through the water to alert other tadpoles to the presence of predators. The researchers found that tadpoles repeatedly exposed to the alarm pheromone over several days showed elevated whole-body levels of corticosterone.

In the laboratory, other tadpoles were exposed either to the alarm pheromone, to corticosterone or to a chemical that blocks the synthesis of the stress hormone. Over the course of several days, tadpoles treated with either the pheromone or the stress hormone developed deeper tails and shorter trunks than control animals, while tadpoles treated with the pheromone and the hormone inhibitor had shallower tails and longer trunks than those exposed to the pheromone alone.

"A key finding was showing that you could eliminate the effect of the alarm pheromone on tadpole body shape by blocking the production of the stress hormone," Denver said. "If you block production of the animal's hormone and it inhibits the change in tail size, then that's a powerful argument that the production of corticosterone is physiologically important for the morphological change."

In another experiment, tadpole tails were placed in a petri dish containing corticosterone. Over the course of several days the tails grew larger, suggesting that the hormone was acting directly on the tail to make it grow.

"The action of the stress hormone on the tail to cause it to grow was unexpected because in adult vertebrates, including humans, prolonged exposure to stress hormones typically inhibits the growth of tissues," Denver said. "In humans, chronic stress causes muscle wasting."

In another set of experiments, normal-tailed tadpoles and large-tailed tadpoles produced by exposure to corticosterone or alarm pheromone were placed in tanks containing uncaged dragonfly larvae, which were allowed to attack the tadpoles. The large-tailed tadpoles had a higher survival rate than their smaller-tailed neighbors.

The third author of the Proceedings of the Royal Society B paper is Earl Werner, director of the E.S. George Reserve and a professor in the U-M Department of Ecology and Evolutionary Biology.

All experiments were conducted in accordance with the guidelines of the University Committee on the Use and Care of Animals at the University of Michigan. The work was supported by the U-M Department of Ecology and Evolutionary Biology and by several grants from the U.S. National Science Foundation.

Prehistoric Wolf Ice-Skated to Remote Island

The mystery surrounding the origin of a wolflike predator that once lived near Antarctica ? a puzzle that stumped even Charles Darwin ? has now been solved, researchers say.

The extinct carnivore apparently made its way to islands hundreds of miles from the nearest continent by crossing the frozen sea thousands of years ago, scientists explained.

The reddish coyote-sized Falkland Islands wolf was the only mammal native to the Falkland Islands far off the east coast of Argentina. The foxlike predator lived on seals, penguins and sea birds until hunters exterminated it in 1876.

The existence of the Falklands wolf perplexed Darwin when he first encountered it in 1834. "How did this great big carnivore arrive to a set of islands 460 kilometers (285 miles) from the nearest mainland when no other terrestrial mammal did?" asked researcher Alan Cooper, an evolutionary biologist at the University of Adelaide in Australia. "If it came by a land bridge, then the islands should've been covered with rodents as well, since South America is rodent central."

"It was incredibly tame ? it swam out to meet sailors, wagging its tail," Cooper told LiveScience. "That led to suggestions that it was a semi-domesticated dog that Native Americans took out while hunting, explaining how it got to the Falklands when there were no other mammals there." However, past analysis of DNA from museum specimens of the Falklands wolf, including one that Darwin collected, revealed it was not a dog after all. Instead, its nearest living ancestor was the maned wolf (Chrysocyon brachyurus) from the South American savannas, an odd predator resembling a red fox with almost stiltlike legs.

To help solve the mystery of how the Falklands wolf colonized the islands, Cooper and his colleagues compared its DNA with remains of what seemed like its closest extinct mainland relative, Dusicyon avus. This carnivore is similar to the Falklands wolf, save for smaller teeth and jaws.

The analysis suggested the Falklands wolf did not become isolated from its mainland cousins until about 16,000 years ago, before scientists think humans arrived this far south in South America. This time coincided with the last height of the ice age, when glaciers covered large portions of the planet.

"The sea levels around the world were really low then, since all the water was frozen onto icecaps ? in fact, they were about 130 meters (425 feet) lower than currently," Cooper said.

The researchers then sleuthed through past research and found references to now-underwater terraces of rock off the coast of Argentina that would have created a narrow, shallow marine strait back when sea levels were lower. "This strait would've been only 20 to 30 kilometers (12 to 18 miles) wide, but when it was frozen over during really cold periods, the Falklands wolf could've effectively walked across the strait to the islands," Cooper said.

Although the ancestors of the Falklands wolf apparently proved more than willing to wander across this bridge, "rodents don't like walking across large amounts of ice," Cooper said, explaining their absence from the islands.

The scientists detailed their findings online March 5 in the journal Nature Communications.

My colleagues at the Room for Debate blog have initiated a valuable discussion around this question:

Fish plays a starring role in the?Mediterranean diet?that?s supposed to keep us healthy. And yet it?s a?disappearing commodity. Meanwhile, there is growing concern about?what is sustainable, or even?identifiable. Is there a way to meet consumer demand for quality seafood and protect threatened marine life at the same time?

The conversation includes two of my favorite fish mavens ? Paul Greenberg, the author of ?Four Fish,??on the role of sustainable aquaculture (a particular focus of mine) and Callum Roberts, the author of??The Ocean of Life: The Fate of Man and the Sea,??on the value of no-fishing zones.

But there are other important contributions, including a push to re-examine the size limits on lobsters in Maine and other states from?Diane Cowan,?a senior scientist at?The Lobster Conservancy:

The intent of a minimum size is to give lobsters a chance to reproduce at least once. Unfortunately, extreme harvest rates targeting one-pound and 1.25-pound lobsters have led to a situation in which size limits fall short of their aim to increase reproductive output. While part of the problem is that harvest rates minimize the number of lobsters growing to sizes exceeding the maximum, the larger problem is that a vast majority of harvested lobsters are too small to have borne offspring. Worst of all, while lobsters can reach sizes exceeding 40 pounds, intense fishing ? targeting lobsters weighing as little as one pound ? has truncated the size structure of the population in favor of small lobsters.

What she says about lobsters applies to many fish species, as well. In both cases, the goal should be to make sure the sea retains plenty of ?BOFFs,? an acronym used by marine conservationist Carl Safina and Jane Lubchenco, the former administrator of the National Oceanic and Atmospheric Administration, to describe the ?big old fat females? that are huge egg producers and the cornerstone of healthy marine species populations.
